Glutamic Acid Has a Liver-Protective Effect through the Suppression of Inducible Nitric Oxide Synthase in Primary Cultured Rat Hepatocytes
نویسندگان
چکیده
Background: Theanine, which is hydrolyzed into glutamic acid (Glu), excerts beneficial effects on organ metabolism. However, there is little scientific evidence that Glu influences inflammatory mediator such as tumor necrosis factor (TNF)-α and nitric oxide (NO).In inflamed liver, proinflammatory cytokines stimulate liver cells, followed by the induction of inducible NO synthase (iNOS). Excessive NO produced by iNOS has been implicated as one of the factors in liver injury. Thus it is essential to inhibit iNOS induction for the prevention of liver injury. In this study, we examined interleukin (IL)-1β-stimulated hepatocytes as a simple “in vitro injury model” to investigate liver protective effects of Glu.

The Effect of Dexamethasone on Expression of Inducible Nitric Oxide Synthase Gene During Liver Warm Ischemia-reperfusion in Rat
Background: Liver ischemia / reperfusion Injury (IRI) is one of the major causes of liver failure during various types of liver surgery, trauma and infections. The present study investigates the effect of dexsamethasone on the liver injury and inducible nitric oxide synthase gene expression during hepatic warm ischemia/reperfusion in rats. Materials and Methods: 24 male Wistar rats (200-250 g)...
متن کاملpyroGlu-Leu inhibits the induction of inducible nitric oxide synthase in interleukin-1β-stimulated primary cultured rat hepatocytes.
Pyroglutamyl leucine (pyroGlu-Leu), which is a peptide isolated from wheat gluten hydrolysate, has been reported to be a hepatoprotective compound in acute liver failure. In inflamed liver, proinflammatory cytokines including interleukin (IL)-1β and tumor necrosis factor (TNF)-α stimulate the induction of inducible nitric oxide synthase (iNOS). Excess production of nitric oxide (NO) by iNOS is ...
متن کاملEffect of dexamethasone on the endothelin-1 (ET-1) and endothelial nitric oxide synthase (eNOS) genes expression during hepatic warm ischemia/reperfusion in rat
Background: Hepatic ischemia/reperfusion injury (I/RI) is a multifactorial pathophysiologic process which can lead to liver damage and dysfunction. This study examined the protective effect of dexamethasone on the gene expression of endothelial nitric oxide synthase (eNOS) and endothelin-1 (ET-1) and on the liver tissue damage during warm hepatic I/R. Materials and Methods: A total of 32 mal...
متن کاملImprovement of Tissue Survival of Skin Flaps by 5α-Reductase Inhibitors: Possible Involvement of Nitric Oxide and Inducible Nitric Oxide Synthase
Background: Skin flap grafting is a popular approach for reconstruction of critical skin and underlying soft tissue injuries. In a previous study, we demonstrated the beneficial effects of two 5α-reductase inhibitors, azelaic acid and finasteride, on tissue survival in a rat model of skin flap grafting. In the current study, we investigated the involvement of nitric oxide and inducible nitric o...
متن کاملThe anti-inflammatory and anti-apoptotic effects of gallic acid against mucosal inflammation- and erosions-induced by gastric ischemia-reperfusion in rats
The present study aimed to evaluate the protective effect of gallic acid on gastric mucosal lesions caused by ischemia-reperfusion (I/R) injury in rat. Forty male rats were randomly divided into sham, control (I/R injury) and three gallic acid-pretreated groups. To induce I/R lesions, the celiac artery was clamped for 30 min and then the clamp was removed to allow reperfusion for 6 hr. Pretreat...
